COX-2 is induced and responsible for synthesizing prostaglandins that are important mediators of pain, inflammation, and fever. However, there are overlapping functions of the mediators derived from these isoforms. Naproxen is a nonselective inhibitor of COX-1 and COX-2. The pharmacokinetics of naproxen in dogs and horses differ substantially from people. Whereas in people the half-life is approximately 12-15 hours, the half-life in dogs is 35-74 hours and in horses is only 4-8 hours, which can lead to toxicity in dogs and brief duration of effects in horses.
